For only a bit more it would be hard to say no to the excellent M2 Comp ($59k, actually has minimal options), which is also more practical. And if you want a true 2-seater it's hard to look past various new and used iterations of the C7.
In all, it's an okay car, but I just would have hoped for more after the gorgeous initial concept and a 27-year wait.

A nicer interior is worth something, I guess it also depends if the Supra is better at performing on the street/track versus the Z. Supra always used to be the benchmark back in the 90's when the RX7, 300 & Supra were all competing against each other. I thought the RX7 looked better than both the Supra and the Z back then, this new Supra looks modern and current.

If you can afford this car, you can afford the M2. Both are built by the same manufacturer, but one has a patently better drivetrain: manual or DCT and stronger motor. Unless you really love the styling or want an automatic, you get the M2 since you're really not getting a supra. You're getting a BMW with a supra wrap. So why not get the better BMW?
I have an open mind regarding the transmission. I'm not going to dismiss it until I try it out.
Engine horsepower at 335 doesn't bother me at all. For sure 400hp will be easy to get.
My main gripe with the car is that 3400 lb. is too heavy. I really hope they bring the 4-cylinder version to the U.S., it should be closer to 3200 lb.

Would you really choose a car based on whether it might be perceived as a "boy toy" or a "man's car"?!
I'm guessing the Supra is going to be a helluva lot more fun to toss around back roads and at the track. Yeah, I'll go with that whether anyone consider's it a "boy toy" or not...
