aftermarket ecu drivable right away ? or tune first?
if i put and aftermarket ecu ( haltech,hondata,aem,ect) in my ca,r can i drive the car down to the tuner after or will the car have to be towed around until it is properly tuned ?

If you buy something like the Karcepts AP1 Emanage setup, then it is plug and play (tune already applied, but you really want the other supporting mods with it) but for the most part, yes, you will need to load base maps before heading to the tuner if you plan on driving it.

Keep in mind it's not usually as simple as just loading a base map and driving down the road. If you aren't using AEM or Haltech or another manufacturer that highly support specifically the S2000, you'll need to define things like trigger inputs for cam/crank sensors. Not such a huge deal, but depending on your ECU, you'll need some general knowledge on how/where to do that along with things rising edge vs falling edge, reluctor type, and so on. And that's just to get the car to start.
Though it really depends on the ECU you choose also. On something like a Platinum Pro or the MS3Pro the dude on here sells, the base map will eliminate a lot of that headache. On a like an off-the-shelf Elite or generic Megasquirt, you'll need to input all that stuff yourself.
Other things that are good to have but not strictly necessary for startup are like a timing light and a wide-band readout. You don't technically need them, but they help you verify that you're starting up ok and eliminate unknowns.

Why are you considering an aftermarket ECU, something wrong with your OE unit? While the '06 and later cars can be Hondata FlashPro tuned on the road others are not so simple and dyno time is very expensive. If I had a pre-2006 car (like your MY 2000) I'd not consider an aftermarket ECU unless I was going with forced induction. And you'll need to be flat-bedded to the dyno to tune these money pits.
