Do you want to do the standard sight seeing?
Do you like museums? Art?
What kind of dining do you like?
Where are you staying?
What have you heard about NY that you would like to experience?
Nighlife could mean clubbing, Times Square, Theatre etc.
Can you be more specific?

Statue of Liberty, Empire State Building, Central Park, go to a Mets game, hit up chinatown, little italy, go visit the Met (Metropolitan Museum of Art) if you're into that kinda thing, tons of clubs in the city...not sure what you're into, plenty of nice restaurants to go eat as well (very $$$ for the most part), my favorite is Del Frisco's...u gotta try the crab cake.
sorry for the run-on sentence...too lazy to make sentences

From Manhattan, take the 4 train (aka muggers' express ) up to 161st Street / Yankee Stadium.
