Route 66: Chicago to L.A.
I just drove from MKE to NYC and back two weekends ago... 17 hours straight each way... don't be afraid to stop often and throw a frisbee or a football around for a bit. As long as you're not going for any speed records taking some time for physical activity does wonders for the back.
I taped up the front of my car as well... I'm sure it saved some rock chips but was more to be ridiculous.

Route 66 trip sounds awesome, I'd love to do one as well
Take as many pics as you possibly can.
Oh, and I agree with Charlie, stop often and take in some sights, it really breaks up the drive and makes it much more enjoyable.

Drove from Jacksonville, FL, to Times Square New York for the Fourth of July in 07, stayed two days, then straight to San Diego from NY. I made the trip alone so a co pilot will help for sure. XM is your friend. Oh, and DO NOT get fuel in Utah, unless you like hiking....
Tape up the car. Tape up the car. Tape up the car.
I went from Madison to Phoenix last summer.(In my celica)
Make sure you got AAA. Make sure the refrigerant is full for the A/C. My trani threw a syncro and the AC went out. Drove the remaining 2000 miles of the trip in 4th gear with No A/C.
Sleep When you need to. Drove there and back a day and a half each time. I do not recommend it. lol.
Take pictures, bring extra memory cards and batteries, or an in car charger.
All i can think of for now. A Road side safety kit wouldnt hurt(flares, flash light, fix-a-flat, etc etc.

I have made this trip in an AP1 and an AP2. Gas in Utah was the only killer in both cars.... Never turned the AC on, it's a convertible. I have cell pics in Death Valley with the outside temp indicator at 109, topdown. Cowboy up!!!
Here is a link to a "things I learned thread" on one of said trips a few years back:

Top Down, AC on.
For really long roadtrips I find the most important thing to have is in ear headphones. (in a convertable or on a motorcycle, either way, very important imnsho)
a DC to AC inverter is good as well for whatever you might need to charge. sun screen.
If I had an unlimited amount of money, and spare time, I would make this trip once a month.
