i've read somewhere that a possible reason why it was important to save the cheerleader was because if the claire was killed, then sylar would had adopted her powers making sylar self regenerative and virtually impossible to kill, and thus "save the cheerleader, save the world"
Isn't Sylar already impossible to kill? The cop shot him a few times but he got up. What else could you reasonably throw @ a guy to kill him? The quiet black d00d already got Sylar by negating his powers.

the power in all the "heroes", according to mohinder's dad, is in the brain. dismantle sylar's brain and i guess you can destroy sylar. it's probably the reason why claire wasn't able to regenerate when she had a wooden stake impaled into her skull. the stake probably interfered with the part of her brain that was responsible for self regeneration.

I think he is a master at putting things back together a certain way. I think he looks at the brain to figure out what to move to make him have thier powers. I don't know how he can change his own brain yet, but that is my theory.
