Single Knock
From Honda Service bulletin:
Axle Pops or Creaks:
’00–02 S2000
On 0002 S2000s, if your customer complains of
hearing a single pop or creak from the rear of the
vehicle when changing direction from forward to
backward and vice versa, the problem may be
axle (driveshaft) movement in the rear hub. To
eliminate the noise, replace the spindle nut on the
affected axle. Apply grease to the surface of the
new spindle nut where it contacts the hub. Torque
the new nut to 298 N·m (220 lb-ft), and stake the
nut.

I didn't think it was that Paul - but as you were going for hub nuts, thought that would be useful info. I've not a clue what it might be from your description, so have left it up to others to advise
No you didn't Ian, but thanks for posting the info
So, to the others who might know ... let me describe it like this, if you had one spoke on a wheel every time it got to say 12 oclock it 'knocks' and does it on every subsequent revolotion

The only thing I wondered was like a stone in tyre or nail - presume you've checked tyre surface? Is it a loud metallic knock or can you describe it a bit more? (know that's hard)
If someone was to tap the rear axle with a small tack hammer every single revolution of the wheel then that would be like the sound. As you say it's difficult to describe a sound I guess once the wheel is off and a look underneath ensues then we might find it ... and HONDA are ordering in the hub nut so I have that as a fall back if I can't see what else the problem might be.
You all don't think it could be the brake calliper or something ... I had an advisory on my rears being worn and needing replacement but ... would you think likely to make that sound ?
Thanks anyway.

Paul, have you tried jacking off the ground, and turning the wheel by hand whilst in neutral. Or whilst on axle stands with someone else with you, sit in car and select gears and listen for the issue.
Could it be a loose wheel nut? Doubt its your calipers too.
