i'd do what others have said... check wheel nuts first, and if they are tight, jack the back end up and spin the wheels manually.

Wheel nuts have been checked they are tight

Will have it on ramps on Saturday and find the issue, to my ear it sounds like the shaft or something has broken and is clipping, will know Saturday and report back, thanks for your help everyone.

It was the hub nut. Had a new one but tightened up the old one to the recommended torque and all seems good.
