No, the car has to be level because the only way you'll know the tranny is properly filled is when it comes pouring out of the fill hole.
I actually put a big leveler (the kind with a bubble) against one of the frame rails to make sure the car is level.

I would say get 4 jackstands and have it leveled before trying to refill. Besides, you really need that space to get the leverage to pull the bolts off, as well as the drain pan.
i have changed my engine oil with my car on the ground, i didnt jack it up or anything...and i did it at 1am in a community parking lot. thank god for led flashlights. 
i dont know about trans fluid though...
