Anything that changes A/F ratios means re-tune.

Honestly, build your motor to where you want it, and tune last. Don't keep adding stuff afterwards, or you'll sink a lot of cash into dyno-tune time.

maybe, now i'm no expert on the subject, but the ecu should relearn the proper mixture of air/fuel if its only small things are added such as intake or exhaust, but FI applications will need a tune bc of different engine management system, UNLESS you are runnin a supercharger under 7 PSI you can use emanage or vafc2...if i'm not mistaken.
