Well, if you get a good set of snow tires, the S2000 will be just fine in the winter.
As for practicality, it's not great, but coming from an MR2, you're already used to that.
If you really need to haul stuff, go rent a van for $30 a day.
Oh, I wouldn't consider the Celica if I were you. The MR2 is a pinnacle of Toyota design and engineering - the Celica just can't match it.
Incidentally, the MR2 is my favorite Toyota, period. (AE86 is number 2).

Mine's a daily and I have no problem, blowing smoke up my co-worker's butt in the parking lot. Acres of Camrys and Accords and I badass black S2 sitting across three spots. It's a sweet way to start the day!
There is enough room for your gear if you pack light and leave the toneau cover at home.

I use mine as a DD. So far the only problem I've had is taking someone to the airport with a suitcase that almost didn't fit. I took the spare tire out and it made it though. If I didn't have friends that will lend me their trucks and SUVs if I need it, I probably would not have bought the car.
Since you use MT during the week, if you don't need to haul big things on the weekends, then I'd say go for it!

mines a pretty much daily driver as well, if you don't need a lot of trunk space, it works out well as a daily car.
I can't think of anything that would be more fun to drive in nearly any price range.

I took mine on a 600 mile weekend trip and was drained by the time I returned. On day trips the windnoise can be tiresome. I love my S2K and have NO regrets buying it. I use mine for a weekender and LOVE short trip around town. The only drawback - don't buy an S2k if you don't want the attention it will draw
The S2000 is at least as practical as the MR2 is. Though the Celica with it's hatchback and rear seats will be more so.
If you get the S2000, get it because it sounds better, handles better, feels better and looks better (IMHO) than the Celica.
