I use it via Bluetooth with my Alpine head unit and iPhone. Do you have a Bluetooth head unit?
Problem with a USB cable is I guess the head unit will want to see your phone as a USB drive with MP3 files in the root directory. Spotify won't work like this, so I would expect the USB solution might not be possible. A 3.5mm jack would work, but you'd want to charge your phone at the same time.

I do have a blue tooth head unit but the hands free is done through it. I've tried it that way but can't seem to get it to play. When i play it it just comes through the speaker on the iPhone.
The aux port I use for an iPod adapter for playing but it's a different connection to my iPod and accessing the back of the head unit isn't easy to unplug and change leads.
